In brief

A lipid-based process, built on shared stocks

A liposome or lipid-nanoparticle (LNP) product is built by dissolving lipids in an organic phase and combining them with an aqueous phase to form vesicles, which are then size-reduced, purified, formulated and filled. From nuclease-free water through hydration and mixing buffers, diafiltration and purification stocks, to excipient-grade formulation, PAN-Biotech delivers the coherent process chemistry that carries a liposome product from lipid stock to the final formulated drug product. A small set of qualified PAN buffers, stock solutions and stabilisers recurs across the whole workflow - one partner, one qualified supply base, from research to GMP.

The challenge

A chain of buffers, stock solutions and excipients, from one supply base

A liposome programme runs on a long chain of buffers, salts, solvents and excipients - from lipid stock and buffer preparation through vesicle formation, size reduction, purification and final formulation. Sourcing them from many suppliers slows qualification and scale-up. PAN-Biotech consolidates the water, buffer, stock-solution and excipient chemistry into one qualified GMP supply base, so a programme can stay with one supplier from research to GMP.

What matters for liposome manufacturing

Buffer, stock-solution and excipient chemistry - from research to GMP

PAN-Biotech covers the water, hydration and mixing buffers, process stocks and excipient-grade formulation chemistry of the liposome process - so a programme can stay with one qualified supplier from first research runs to GMP manufacturing.

Water as the base

WFI Beyond - metal-controlled, ultra-low endotoxin and nuclease-free - is the aqueous base of every buffer, hydration, exchange and formulation step.

Hydration & loading buffers

Phosphate, HEPES, Tris and neutral citrate for hydration at physiological pH, plus ammonium sulfate stocks for remote-loading gradients.

LNP mixing buffers

Acidic acetate and citrate buffers for the aqueous phase of microfluidic mixing, with Acetic acid 1 M and NaOH 1 M for pH adjustment.

Excipient-grade formulation

Histidine, phosphate and Tris buffers, DPBS and 0.9 % saline, polysorbates, sucrose, trehalose, mannitol, glycerol, L-arginine and rHSA - the excipient chemistry of the final product.

The stock-solution principle

A handful of stocks, reused across the process

Rather than a separate finished buffer for every step, the liposome process is built on a few concentrated stock solutions that recur across the workflow - one qualified stock covers several steps, so there are fewer SKUs to qualify and full freedom to prepare working buffers to your own recipe. How often each recurs:

Quality & regulatory

The GMP and regulatory foundation

Most products named here are manufactured by PAN-Celltech and available at GMP grade; the buffers, salts, stock solutions and formulation excipients carry a CT article number and are available directly, several also with a research-grade (RUO, P-) equivalent. Two are research grade only: PANclear Octoxynol and the RNase/DNase Neutralizer. Two of the most-used utilities go further: DPBS (CT-36500) and WFI Beyond are backed by filed FDA Drug Master Files that customers can reference in their own submissions. And we go one step further: on a customer's request, we will file the PAN products they use with the FDA as well. A DMF-listed product makes their documentation and approval process faster and easier - the same regulatory shortcut we already provide for DPBS and WFI Beyond, extended to the products that matter most to them. Manufactured to GMP in an EU Annex 1 facility within an ISO 9001 quality system - audit-proven and GMP-compliant, with GMP compliance confirmed in customer audits. One qualified stock covers several steps; a fully customized GMP formulation is available at any step, to your recipe, without switching supplier.

The water underneath

Beyond compendial, on request

Water is the base of every buffer and stock, so it sets the floor for the whole process. Alongside standard WFI quality water, PAN offers WFI Beyond - Water for Injection engineered beyond the Ph. Eur. / USP monograph and backed by its own filed Drug Master File. Every downstream buffer on this page is available in a beyond-compendial version produced with WFI Beyond, on request, for processes with the most demanding endotoxin and purity requirements - one qualified foundation, reused everywhere.

Questions

Frequently asked questions

Which buffers do I need for liposome manufacturing?
PAN supplies the aqueous and exchange buffers across the whole train - hydration buffers (ammonium sulfate, HEPES, phosphate, acetate, Tris, citrate), diafiltration and saline solutions, high-salt and EDTA purification buffers, and formulation buffers (histidine, phosphate, Tris) - all built on WFI quality water or WFI Beyond.
Do you supply the lipids or the organic solvents?
PAN supplies the process liquids - buffers, stock solutions, cryoprotectants and water - not the lipids themselves. The lipids and the organic solvent (typically absolute ethanol) for lipid stock solutions and microfluidic mixing are customer-supplied.
What is available for formulation and stabilisation?
Cryoprotectants (sucrose, trehalose), stabilisers (mannitol, glycerol, L-arginine, recombinant HSA), surfactants (polysorbate 20/80, at low concentration where compatible with the bilayer), formulation buffers (histidine, phosphate, Tris, DPBS) and tonicity agents (saline, dextrose) - all on WFI Beyond.
Can every buffer be supplied beyond compendial?
Yes. Every aqueous buffer can be produced on the beyond-compendial WFI Beyond water base instead of standard WFI - metal-controlled and nuclease-free, backed by a filed FDA Type IV (excipient) DMF.
Is the toolkit GMP grade?
The downstream buffers and stock solutions are available in GMP grade (CT-series) with a CoA per lot; several are also available as catalogue (RUO, P-series) products. Custom GMP buffers to your recipe are available on request.
