Erythrocyte Lysis Buffer (1x) is a ready-to-use buffer for the selective lysis of red blood cells in samples containing white blood cells, such as EDTA-, citrate- or heparin-treated whole blood, buffy coats and bone marrow. It contains ammonium chloride, potassium hydrogen carbonate and EDTA, is sterile-filtered (0.2 µm) and contains no fixative and no preservatives. The ammonium chloride lyses red cells with minimal effect on lymphocytes when used as directed.

Add lysing buffer to whole blood (e.g. 8-10 ml per 1 ml blood), incubate ~15 min at room temperature, centrifuge and wash the leukocyte pellet.
