Dispase II is a neutral metalloprotease from Bacillus polymyxa, supplied at 2.4 U/ml in DPBS without Ca and Mg. This purified, animal-origin-free protease hydrolyses N-terminal peptide bonds of non-polar amino acids and acts mildly, which makes it gentle on cell membranes and well suited to preparing primary cells and to subcultivation. It is commonly used to separate epidermis from dermis, for stem cell and hepatocyte isolation, and as a secondary enzyme alongside collagenase.
