Trypsin 0.05 % / EDTA 4 Na 0.02 % in HBSS, with Phenol red is a ready-to-use cell detachment solution containing 0.05 % trypsin and 0.02 % EDTA (4 Na), formulated in HBSS without calcium and magnesium, with phenol red. Trypsin is a porcine pancreas-derived enzyme (1:250) that dissociates anchorage-dependent cells; EDTA is added to chelate calcium and magnesium and enhance trypsin activity. A low-concentration trypsin-EDTA on an HBSS base with the tetrasodium EDTA salt and phenol red indicator. After detachment, trypsin should be neutralised with serum-containing medium or a trypsin inhibitor.
