(10x) Trypsin 0.5 % / EDTA 0.2 % in DPBS, w/o Ca & Mg is a ready-to-use cell detachment solution containing 0.5 % trypsin and 0.2 % EDTA, formulated in DPBS without calcium and magnesium. Trypsin is a porcine pancreas-derived enzyme (1:250) that dissociates anchorage-dependent cells; EDTA is added to chelate calcium and magnesium and enhance trypsin activity. This is a 10x concentrate: dilute 1:10 with Ca/Mg-free DPBS to obtain a 1x working solution. After detachment, trypsin should be neutralised with serum-containing medium or a trypsin inhibitor.

Dilute the 10x concentrate 1:10 with DPBS (w/o Ca, Mg) before use. Aspirate medium, rinse the monolayer with Ca/Mg-free salt solution, add enough pre-warmed trypsin to cover the cells and incubate until they round up and detach; then neutralise, centrifuge and resuspend in fresh medium.
