Trypsin 0.25 % in DPBS, w/o Ca & Mg is a ready-to-use cell detachment solution containing 0.25 % trypsin, formulated in DPBS without calcium and magnesium. Trypsin is a porcine pancreas-derived enzyme (1:250) that dissociates anchorage-dependent cells; this variant is supplied without EDTA, for applications where chelation is not desired. It is chosen when a pure trypsin action is preferred over a trypsin-EDTA combination. After detachment, trypsin should be neutralised with serum-containing medium or a trypsin inhibitor.

Aspirate medium, rinse the monolayer with Ca/Mg-free salt solution, add enough pre-warmed trypsin to cover the cells and incubate until they round up and detach; then neutralise, centrifuge and resuspend in fresh medium.
